So I already picked up my invites. My initial time was 4pm which is printed on the invitations but now my matron of honor, who is my sister tells me she has a test the day of the wedding and it's 2 hours away from our city. Her test is at 1 pm and she thinks she can make it back in time. I plan on changing the time to 5 pm but how should I do it? Should I white out the time and write it in or should I include a note in the invitation about the time change?

Your sister should tell her professor she will be out of town and needs to take the test at an earlier date.

I once had a weekend seminar that conflicted with a final exam and the professor allowed me to come in for a make-up at an alternative time. I think an earlier date is preferable to a later date in case there are concerns about cheating or anything like that. (Eta: spelling)
