Where do you live and how far do you want to drive? The Midwest is big.

We went on a weekend trip to Chicago a few years ago and it was a blast. However one of the most fun things we did was go to the cubs game and you will be too late in the year for that.

Door county is also a great more low key place. The north shore is great, Duluth or more north up by the gunflint trail. But it depends what you like to do, you will be too late in the year for water sports but too early for skiing.

St Louis is awesome and there are a ton of great places nearby for more relaxation.

St Genevieve/Hermann MO are great small towns with a fabulous wine trail and it is only an hour drive to St Louis City.

Pere Marquette State Park in Grafton, IL is also highly recommended. Beautiful park, great lodge. Great small town with stuff to do, but it is less than an hour to St Louis City. We frequently stay there for weekend getaways. They have basic hotel type rooms in the lodge, but there are also amazing cabins with fireplaces. The fall foliage is beautiful for pretty much the entire month of October. There are really amazing views that you can either hike or drive to. Several wineries (including one with a zip line from the bluff to the river, if you are adventurous) are just a few minutes drive. There are events and fall festivals all over the area that can be a ton of fun.
